老化影响内源性干细胞的功能和数量。特别是,在老化过程中,凋亡细胞死亡和衰老的增多可以降低其分化多样性。此外,老化和老化相关疾病影响了干细胞的旁分泌活动和他们的移植的效率。总的来说,这将导致器官自我修复能力的减弱,原因可能是由于降低了干细胞的功能。因此,本研究专注于通过过度表达抗衰老和抗凋亡基因来提高处于衰老期干细胞的修复能力。在本文中,我们描述了干细胞衰老过程中的关键基因和信号通路,并讨论能够使干细胞恢复活力体外的基因改造方法,为心血管系统疾病的治疗提供重要依据。

摘要: Aging affects endogenous stem cells in terms of functionality and numbers. In particular, during aging, the stemness property can decrease because of enhanced apoptotic cell death and senescence. In addition, aging and agingrelated co-morbidities affect the paracrine activity of stem cells and the efficiency of their transplantation. Collectively, this leads to a reduction of the capacity of organs to repair themselves, possibly due to a reduced functional capability of stem cells. Therefore, major efforts have been invested to improve the repair capability of stem cells in aged individuals by overexpressing antisenescence and antiapoptotic genes. In this review, we describe critical genes and signaling pathways in stem cell aging and discuss ex vivo genetic modification approaches aimed at stem cell rejuvenation that are of interest for the cardiovascular system.
