Experimental Study of Therapeutic Efficacy of the Topical Preparation «Kourochitin» in Skin Allergy

Abstract:

Aims:The manuskript aims to demonstrate the results of study of dermatoprotective action a new topical preparation "Kourohitin", which was developed on the base of quinazoline alkaloid tryptanthrin by researchers of PIBOC RAS.

Background: As well known quinazoline alkaloid tryptanthrin posses to anti-inflammatory, antibacterial and anti-allergic activity, and exhibiter a therapeutic effect in the treatment of allergic skin desises. PIBOC RAS have the possibility to be obtained the tryptanthrin with good yields by different synthetic methods. The researchers of PIBOC RAS have developed the new tryptanthrin-containing topical preparation called "Kourohitin" which can act as anti-inflammatory, anti-allergic, and wound healing remedy.

Objective: Evaluation anti-inflammatory, anti-allergic, and wound healing action of tryptanthrin-containing topical preparation "Kourohitin" in using the experimental murine models of skin allergy and in veterinary research.

Method: Experimental modeling of allergic contact dermatitis, Experimental modeling imiquimod-induced psoriasis, Evaluation efficacy of «Kourochitin» in treatment of canine atopic dermatitis in Veterinary research.

Result: In a murine model of 2, 4- dinitrofluorobenzene-induced allergic contact dermatitis (ACD) was show that «Kourochitin» 1) contribute to reduce the level of erythema (hyperemia, lichenification, hemorrhage and tissue necrosis areas) in the allergen damaged skin area and increases the healing index of the epidermis; 2) normalizes the content of eosinophils, basophils and monocytes in the blood of experimental animals; 3) inhibits the production of main pro-inflammatory cytokines: interleukins – 1 and 2, interferon-gamma, and granulocyte-macrophage colony-stimulating factor. In a murine model of imiquimod-induced psoriasis was show that «Kourochitin» application led to reduce a psoriasis severity on inflamed epidermis of experimental animals. Moreover, in veterinary research «Kourochitin»-treatment of canine atopic dermatitis almost completely eliminated signs of allergic manifestations in the epidermis.

Conclusion: Based on literature and experimental data it can be assert that "Kourohitin" is the dermatoprotective topical remedy with wide range of pharmacological activity, that is having anti-inflammatory, wound healing and antiallergic action. Therefore, this drug can be widely used for therapy of various dermatological diseases, mainly allergic skin lesions.

Other: In the present work it shown, that «Kourochitin» as a topical preparation can be successfully used to treat dermatological diseases, particularly allergic skin lesions.
