Antioxidant Activities of N-ferrocenylmethyl-2- and -3-nitroaniline and Determination of their Binding Parameters with Superoxide Anion Radicals

Abstract

Introduction: The antioxidant activity of two potential antioxidants N-ferrocenylmethyl-2- nitroaniline (2FMNA) and N-ferrocenylmethyl-3-nitroaniline (3FMNA) was evaluated using spectrophotometrical and electrochemical technics.

Methods: Spectrophotometrical assays were carried out using stable 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH) and electrochemical assays were based on measuring the oxidation peak current of relatively stable superoxide anion radical O2˙¯ electrochemically generated by reduction of commercial molecular oxygen in DMF.

Results: 2FMNA shows highest antioxidant activity in both methods DPPH and O2˙¯ radicals scavenging activities (4.27 ± 0.01mM) and (9.38± 0.09 mM) respectively which is almost equal to that of standard antioxidant α-tocopherol (4.81± 0.03 mM) and (9.50± 0.09 mM). The diffusion coefficients of the free and O2˙¯ bound forms of 2FMNA and 3FMNA were evaluated from Randles–Sevcik equation. Cyclic voltammetry based assay was developed for the calculation of binding parameters like binding constant, ratio of binding constants and binding free energy.
