摘要
癌症被认为是美国的主要死因之一。虽然已经制定了预防策略,早期发现和改进的治疗方案,但仍需要新的靶点和治疗方法。自从得出结论认为癌症是由细胞的遗传和表观遗传改变介导的,许多研究小组正在关注通过营养,表观遗传机制和非编码RNAs预防和治疗的其他手段,这些手段已被证明可以控制基因表达,不同的功能在细胞水平。随着人类癌症中高通量测序的出现,鉴定疾病的新型生物标志物和治疗靶标的潜力已经大大增加,并导致许多在各种癌症中失调的非编码RNA的鉴定。基因表达和调节对于维持正常组织和细胞的稳态是重要的。并不罕见,由于转录靶标的表达增加或减少,特定基因的上调或下调与癌症相关。本综述重点介绍了营养在癌症中的作用以及非编码RNA的失调,特别强调了长链非编码RNA和不同癌症类型的microRNA。

摘要: Cancer is considered one of the leading causes of death in the United States. Although preventive strategies, early detection, and improved treatment options have been developed, novel targets and therapeutics are still needed. Since concluding that cancer is mediated by genetic and epigenetic alterations of the cell, many research groups are now focusing on other means of prevention and therapy via nutrition, epigenetic mechanisms, and non-coding RNAs which have been shown to control gene expression and have many different functions at the cellular level. With the advent of high-throughput sequencing in human cancer, the potential to identify novel biomarkers and therapeutic targets of disease has increased tremendously and led to the identification of many non-coding RNAs that are dysregulated in various cancers. Gene expression and regulation is important in maintaining the homeostasis of normal tissues and cells. Not uncommonly, up- or down-regulation of particular genes are associated with cancer as a result of increased or decreased expression of transcriptional targets. This review focuses on the role of nutrition in cancer and the dysregulation of non-coding RNAs with particular emphasis on long non-coding RNAs and microRNAs in different cancer types.
