摘要

靶向高度增殖的细胞是许多类型的侵袭性肿瘤的重要问题。 增殖细胞核抗原(PCNA)是一种必需蛋白,它参与DNA代谢的各种过程,包括DNA复制和修复,染色质组织和转录以及姐妹染色单体凝聚。 另外,PCNA参与细胞存活,并可能参与能量代谢的途径,例如糖酵解。 因此,正在积极研究针对这种蛋白质靶向化学疗法以对抗高度增殖的恶性肿瘤的可能性。 当前,用靶向PCNA的药物治疗细胞的方法依赖于小分子或与PCNA结合或充当相互作用伴侣竞争者的肽的使用。 在这里,我们描述了靶向PCNA的药物开发中的技术现状,并讨论了它们在不同类型的肿瘤细胞系和动物模型系统中的应用。
