Abstract:A new, simple, accurate and precise HPLC-DAD method has been developed and validated for quantitative
determination of solasodine in aerial parts of Solanum nigrum Linn. The column used was C18 (150 mm X 4.6 mm I.D., 5
μm) and mobile phase used was acetonitrile: 10 mM ammonium acetate buffer pH 7.7(65:35,v/v).The flow rate of mobile
phase and detection wavelength were kept at 2.0 ml/ min and at 205 nm, respectively. The column oven temperature was
set at 40 °C. The linearity of the method was found over a range of 300-1500 µg/ml with LOD and LOQ of 100 µg/ml and
300 µg/ml, respectively. The method has shown recoveries of solasodine in the range from 100.12 % -106.67 %, with low
intra and inter-day % R.S.D. (0.89% and 1.25%, respectively). The method was successfully applied for quantitative
evaluation of three extracts and different Ayurvedic formulations for the content of solasodine.
