导致神经退行性疾病和癌症细胞死亡发展的通路可能有重叠,有趣的是,主要的神经退行性疾病诱导因子被证明在癌症增长的调节中起着十分重要的作用,神经退行和癌症中另一个常见的因素是硝基的氧化应激。取决于不同的环境因素,硝基氧化应激可能导致癌细胞再生或者死亡。另一方面,它可能导致许多的神经退行性疾病。 基于文献数据,我们认为甲氧雌二醇在神经退行性发展以及癌细胞死亡之间存在着生理联系,甲氧雌二醇是一个生理化合物,是 17β-雌二醇的代谢物。有趣的是,甲氧雌二醇,商标名为Panzem,也被当作专利抗癌剂,目前在一些临床试验中被调研,本文章旨在探索关于抗癌和甲氧雌二醇的神经毒性,这些有可能会对癌症病人产生长期的药物神经副作用。

摘要: The pathways leading to development of neurodegenerative disease and cancer cell death may overlap. Interestingly, the main neurodegeneration-causing factors have been suggested to play a pivotal role in the regulation of tumor growth. Another common component in neurodegenerations and cancer is nitro-oxidative stress. Depending on the environmental conditions, the nitro-oxidative stress may lead to cancerogenesis or cancer cell death. On the other side, it may also result in a variety of neurodegenerative diseases.
Based on the literature data, we suggest that 2-methoxyestradiol may be considered as a physiological link between development of neurodegenerations and cancer cell death. 2-methoxyestradiol is a physiological compound, a metabolite of 17β-estradiol. Interestingly, 2-methoxyestradiol, branded as Panzem, is also considered as a potent anticancer agent, currently investigated in several clinical trials. The review aims on making closer the facts concerning anticancer and plausible neurotoxic activities of 2-methoxyestradiol, which may suggest adverse neurological long-term effects of the drug in cancer patients.
