目前,细菌耐药性,尤其是对最常用抗生素的耐药性已被证明是一个严重的医疗问题。医院和社区获得性感染通常由耐多药菌株引起。因此,我们被迫开发替代或支持疗法来成功治愈危及生命的感染。使用噬菌体等自然细菌的想法已众所周知。许多文章已被发表来证明动物模型以及临床中测试的裂解性噬菌体的高抗菌功效。研究人员还调查了非裂解性噬菌体和温和噬菌体的应用,获得了良好成效。此外,分子生物学的发展和新一代测序方法在工程化基因设计和噬菌体衍生蛋白中开启了新的可能性。鼓舞人心的表现为参与负责细菌包膜退化的病毒感染第一步的噬菌体酶,名为解聚酶。至少有五大这种酶群体—肽聚糖水解酶、endosialidases、endorhamnosidases、海藻酸盐裂解酶和透明质酸裂解酶—具有应用潜力。也对噬菌体裂解周期中负责后代释放的裂解盒基因 (holins, endolysins, spanins) 编码的蛋白感兴趣。总之,本文讨论了关于噬菌体和噬菌体衍生蛋白在临床、诊断和生物技术中的一些问题。

摘要: Currently, the bacterial resistance, especially to most commonly used antibiotics has proved to be a severe therapeutic problem. Nosocomial and community-acquired infections are usually caused by multidrug resistant strains. Therefore, we are forced to develop an alternative or supportive treatment for successful cure of life-threatening infections. The idea of using natural bacterial pathogens such as bacteriophages is already well known. Many papers have been published proving the high antibacterial efficacy of lytic phages tested in animal models as well as in the clinic. Researchers have also investigated the application of non-lytic phages and temperate phages, with promising results. Moreover, the development of molecular biology and novel generation methods of sequencing has opened up new possibilities in the design of engineered phages and recombinant phage-derived proteins. Encouraging performances were noted especially for phage enzymes involved in the first step of viral infection responsible for bacterial envelope degradation, named depolymerases. There are at least five major groups of such enzymes – peptidoglycan hydrolases, endosialidases, endorhamnosidases, alginate lyases and hyaluronate lyases – that have application potential. There is also much interest in proteins encoded by lysis cassette genes (holins, endolysins, spanins) responsible for progeny release during the phage lytic cycle. In this review, we discuss several issues of phage and phage-derived protein application approaches in therapy, diagnostics and biotechnology in general.
