摘要
恶性肿瘤的常规疗法例如化疗和放疗的存活率很低,这是由于细胞对抗癌药物的耐药性以及抗癌药物缺乏靶向定位能力, 导致对健康细胞造成不必要的副作用,迫使治疗剂量的降低,从而随之使得疗效降低。基因疗法采用不同的病毒和非病毒载体来传输感兴趣的基因和促进理想的治疗性蛋白的产生。尽管癌症细胞特异性的转基因投递仍然存在一些需要解决的挑战,但是基因疗法具有治疗作用的特异性蛋白的高水平表达,以及低的脱靶效应,使得其在癌症治疗方面存在巨大的前景。根据潜在的治疗靶标基因的不同,癌症基因治疗可以分为肿瘤抑制基因替代疗法、免疫基因治疗和酶或前体药物基础疗法。本综述将阐明各种癌症细胞中存在的潜在治疗基因投递在体外实验和利用各种病毒和非病毒载体的动物模型方面的一些当前研究进展。

摘要: Conventional therapies for malignant cancer such as chemotherapy and radiotherapy are associated with poor survival rates owing to the development of cellular resistance to cancer drugs and the lack of targetability, resulting in unwanted adverse effects on healthy cells and necessitating the lowering of therapeutic dose with consequential lower efficacy of the treatment. Gene therapy employing different types of viral and non-viral carriers to transport gene(s) of interest and facilitating production of the desirable therapeutic protein(s) has tremendous prospects in cancer treatments due to the high-level of specificity in therapeutic action of the expressed protein(s) with diminished off-target effects, although cancer cell-specific delivery of transgene(s) still poses some challenges to be addressed. Depending on the potential therapeutic target genes, cancer gene therapy could be categorized into tumor suppressor gene replacement therapy, immune gene therapy and enzyme- or prodrug-based therapy. This review would shed light on the current progress of delivery of potentially therapeutic genes into various cancer cells in vitro and animal models utilizing a variety of viral and non-viral vectors.
